抄録/ポイント： シナプス前コリン作動性ニューロンマーカーである小胞性アセチルコリン輸送体(VACh)は,アルツハイマー病(AD)のような認知低下を伴う神経変性疾患の早期診断のためのイメージング剤の開発のための潜在的な内部分子標的である。ベサミコールは高親和性でVAChに結合することが報告されているので,コリン作動性神経障害の診断のためのVAChイメージング剤として多くのベサミコール類似体が研究されている。しかしながら,多くのベサミコール類似体は,VAChの他にシグマ受容体(σ_1とσ_2)に結合するため,ほとんど全てのベサミコール類似体は臨床試験に適していないことが示されている。本報告では,これらの開発されたベサミコール類似体の化学構造と生物学的特性との関係を,特にin vitro結合プロファイルとin vivo領域脳蓄積について調べた。Copyright 2018 Kazuma Ogawa and Kazuhiro Shiba.
