抄録/ポイント： ぶどう膜炎は,世界中の失明のほぼ10%を占める眼内炎症性疾患である。NF-κBシグナル伝達は,炎症性疾患において重要な役割を果たIKK-βの選択的遮断を介してNF-κBシグナル伝達を阻害するIMD-0354は,いくつかの眼疾患モデルにおいて炎症を抑制することを報告した。ここでは,ヒトにおける内因性ぶどう膜炎に対する良く確立された動物モデル,実験的自己免疫性ぶどう膜炎(EAU)モデルにおけるIMD-0354の治療効果を検討した。IMD-0354の全身投与は,EAUマウスにおいて,臨床的および組織学的重症度,炎症性浮腫,およびNF-κB p65の網膜核への移行を有意に抑制した。さらに,IMD-0354処理は,in vitroでいくつかのTh1/Th17仲介炎症誘発性サイトカインのレベルを有意に阻害した。IKD-0354によるIKKβの阻害はマウスEAUモデルにおける炎症反応を改善し,IMD-0354がヒト内因性ぶどう膜炎に対する有望な治療薬であることを示唆した。Copyright 2020 Elsevier B.V., Amsterdam.
