The development of a recombinant protein expression system at low temperature

Research overview:
As a post-genome sequence project, it is very important to characterize known and unknown proteins revealed by sequence analyses. For functional and structural characterization of proteins, recombinant protein expression systems are very useful for obtaining substantial amount of proteins. While several recombinant protein expression systems by mean of various cells as a host have been developed, no system is available for expression of toxic proteins that artificially retard cell cycle progression of the host cell. We assumed that the toxicity of proteins is caused by growth conditions; the temperature for cell growth overlaps with that for functional activity of recombinant proteins and active recombinant proteins spontaneously function in the host cell. Therefore, we are currently developing a recombinant protein expression system at low temperature by use of bacteria cells as a host that can grow a wide rage of temperature of 4-42 degree C.
