Studies on chromosome band structures at the DNA sequence level

Research overview:
The nuclear genomes of vertebrates consist of long compositionally homogeneous GC levels DNA segments (>300kb), GC-poorand GC-rich segments largely correspond to the DNA of G-bands and R-bands, respectively. I am interested in the regionofthe border of G-and R-bands. The border of chromosomal bands tend to be fragile sites or recombinational hot spots. Ourlaboratory showed that the border of chromosomal bands exists in the human major histocompatibility complex (HLA) cluster spanning about 3Mb on chromosome 6 by computer analysis previously. Namely, class II region is GC-poor, while class III and class I regions are GC-rich. By cosmid walking, wehave cloned the entire 300kb region from HLA class II DR .alpha. gene to HLA class III 21-hydroxylase gene. I am planning to determine the structure of G-band and R-band border.
