【請求項1】 突出末端を有する2本鎖DNA(a)と、前記突出末端と相補的な突出末端を有し、かつ更にその突出先端または切欠き端に、前記(a)の切欠き端または突出先端に存在する塩基とは異種の余分な塩基が存在する2本鎖DNA(b)とを、DNAリガーゼを用いて結合させることにより、前記余分な塩基に対応する位置に存在する(a)の塩基を除去して接合させることを特徴とする、DNAの塩基置換方法。
