抄録/ポイント： 【目的】日本住血吸虫の熱ショック因子(SJ-HSF)をクローン化し,同定し,その構造的特徴と可変型のサブタイプを分析する。方法:日本住血吸虫セルカリアをニュージーランド白ウサギに感染させ、42日後ににを行い、流により雌虫、雄虫及び肝臓の肝臓を採集し、TRIZOL法により総RNAを抽出した。RT-PCRを用いて,日本住血吸虫の雌,雄,および卵の全RNAを増幅し,SJ-HSFのオープンリーディングフレーム(ORF)とSJ-HSF可変領域のフラグメントを得て,クローン化,制限酵素消化,および配列決定を行った。DNAMAN8.0,,,およびMEGA6を用いて,日本住血吸虫と住血吸虫住血吸虫のゲノムデータベースとGENBANKデータベースを分析した。SJ-HSFの遺伝子構造特性,機能ドメイン,可変せん断パターン及び異なる種間のHSF配列の相同性と進化樹を解析した。【結果】自己,雄,および虫卵拡増から を得て,陽性組換えプラスミドを得た。SJ-HSF ORFのPBSJHSFF-F,PBSJHSFF-M,PBSJHSFF-E及び可変-F,PBSJHSFS-M,PBSJHSFS-E,制限酵素及び配列決定は正確であった。配列解析により,SJ-HSFは3つの異なるサブタイプを持つことを示した。SJ-HSF-ISOFORM1(2BP),SJ-HSF-ISOFORM2(2BP),SJ-HSF-ISOFORM3(2BP)であった。GENBANKの登録番号はそれぞれKUKU、KX119143、KX119144であり、いずれも日本住血吸虫ゲノム遺伝子-S000780に位置し、8つのエクソンがあり、雌虫、雄虫と卵の段階で3つの亜型が存在した。SJ-HSF-ISOFORM1の発現は高かった。SJ-HSF-ISOFORM1(671 AA)、SJ-HSF-ISOFORM2(683 AA)は、HSF特徴性DBD(DNA BINDING DOMAIN)、HR-A/BとHR-Cドメインを有する。SJ-HSF-ISOFORM3(282 AA)は早期に停止し,HR-Cドメインはなかった。進化ツリー解析により,SJ-HSFの3つのタイプは,HSF1に属し,そして,それは,住血吸虫のHSF源と類似していた。結論:日本住血吸虫の雌、雄虫、卵の段階には3種類のせん断タイプのHSFが存在し、SJ-HSF-ISOFORM1の発現量が高い。本研究はSJ-HSFのSJ-HSPSの防御機構の分子機構の更なる解明のための基礎を提供した。Data from the ScienceChina, LCAS.
