マウスint-1プロトオンコジーンの標的化破壊は中脳及び小脳の発達の重大な異常にいたるSMIマウスはい由来幹細胞の2か所のint-1対立遺伝子の一方を破壊し,変異遺伝子を子孫へ伝えるキメラマウスを作成した。ヘテロ接合性マウスは正常だったが,int-1^-のホモ接合性マウス(I)は,出生前死亡から重大な失調を伴う生存の範囲の表現型を示した。Iのはい形成時の検査により,int-1蛋白質は中脳及び小脳の誘導に重要な役割を果すことが判明した
