【請求項1】 無細胞タンパク質合成系に、 (1)タンパク質をコードする遺伝子、 (2)改変型tRNATyr、および (3)前記改変型tRNATyrをチロシンアナログ化することができるD321Rの変異を有する変異型チロシルtRNA合成酵素 を添加して、前記タンパク質中の特定部位のアミノ酸を非天然アミノ酸に変異させた非天然アミノ酸含有タンパク質を製造する方法。
