Research overview：
大腸菌を宿主とする組み換えタンパク質の大量発現系では、封入体形成が大きな問題となる場合が多い。本計画では、大腸菌内の封入体形成がタンパク質の熱安定性と溶解性の両者に由来する現象であることを踏まえて、現在までの研究で開発してきたタンパク質・酵素の溶解性を著しく向上させる短いペプチド系溶解性向上タグ(特願2007-303780号)とCspAプロモーターによる低温発現系を組み合わせた強力な封入体形成防止法の開発を提案する。
