【請求項1】 ヒトm-カルパインの活性サブユニットタンパク質をコードする遺伝子と、ヒトm-カルパインの制御サブユニットタンパク質のGR領域中の2ヶ所のグリシンリピートの一方又は両方が欠失した改変制御サブユニットタンパク質をコードする遺伝子とが、共に宿主細胞内で発現可能に導入された形質転換宿主細胞を、18°C以上、25°C未満の低温条件下で培養し、得られた培養物から組換えヒトm-カルパインを採取することを含む、組換えヒトm-カルパインの調製方法であって、該宿主細胞が大腸菌である、前記方法。
